Route 666 is a community of underground homeless in New York City. They inhabit the area under Central Park. The leader of the community is a man named Mephisto.

The Route 666 Community has grown to include the members of other smaller communities like the Ho Chi Minh Trail, comprised mostly of former Vietnam War veterans, and the Blockhouse. The community has access to running water and electricity by illegally tapping city infrastructure. Members of the community known as "runners" go to the surface periodically to collect welfare checks, redeem recyclables for money, and to acquire food and drugs.

Known members of the Route 666 Community include:

  • Mephisto, mayor
  • Tail Gunner, chief runner
  • Little Harry, lieutenant—Little Harry was a petty thief and former heroin addict who joined the community after a stint at Attica Correctional Facility.
  • Boy Alice, lieutenant—a former prep school English teacher in Connecticut, Boy Alice turned to alcohol after a divorce left him jobless and broke. He heard about Route 666 through the Manhattan homeless shelters and soup kitchens.
  • Flint
  • Joe Atcitty (deceased)—Joe Atcitty was the first of the Route 666 murders. His headless body was found dumped outside of the Blockhouse almost a year before Pamela Wisher's body was discovered.
  • Fat Boy (deceased)
  • Hector (deceased)
  • Dark Annie (deceased)
  • Master Sergeant (deceased)

After seeing an article in the New York Post about the murder of Pamela Wisher, Mephisto sent Tail Gunner to contact the article's author, Bill Smithback, to arrange a meeting. Smithback followed the runner into the upper levels of the tunnels and spoke to Mephisto through a hole in the floor. Mephisto told Smithback about the rash of murders plaguing the community and warned him about the mysterious Wrinklers. He lamented the city's reaction to Wisher's murder in contrast to their apathy at the identical killing of several of their community, and asked for Smithback's help in bringing attention to their plight.

Later, Special Agent Pendergast and Lieutenant Vincent D'Agosta visit Route 666 posing as the leader of another community and his assistant. They meet several members of the community and are invited to eat "track rabbit" (rat).

Most of Route 666 was "rousted" in anticipation of the flooding of tunnels beneath Central Park in an attempt to drown the Wrinklers. They, along with other homeless communities beneath the park, emerged in the midst of the final "Take Back Our City" rally, violently clashing with protestors after vandalizing nearby storefronts.

The fate of the community following Mephisto's death in the Astor Tunnels is unknown.
